The preservative 4-chloro-m-cresol (4CmC) is a potent and highly specific activator of ryanodine receptor-mediated Ca2+ release and induces contractures in [micro sign]molar ranges in skeletal muscles of Malignant Hyperthermia susceptible (MHS) patients.
Clinical formulas for heparin contain 4CmC in a reasonable amount.
We studied whether these heparin formulas induce contractures in isolated MHS human skeletal muscles and whether significant serum levels of 4CmC are reached after heparinization in cardiopulmonary bypass patients.

METHODS: Muscle biopsy and in vitro contracture test were performed acc. to the European protocol.
With informed consent surplus muscle strips of MHS and normal (MHN) patients were exposed in the same experimental setting to clinical formulas of heparin, i.e. Thrombophob[registered sign] (1mg/ml 4CmC) and Liquemin[registered sign] (0.8mg/ml 4CmC), and to 4CmC (1mg/ml) and preservative-free heparin in increasing concentrations resp..
The effects on muscle contractures were compared between MHS and MHN.
In a second step, serum levels of 4CmC in cardiopulmonary bypass patients were measured after heparinization (500U/kg) by HPLC at 5, 10 and 60 min.
Statistical analysis was performed by U-test (p < 0.05).

RESULTS: 7 MHN/3 MHS muscle strips were incubated with Thrombophob[registered sign], 9 MHN/6 MHS with Liquemin[registered sign], 6 MHN/7 MHS with 4CmC, and 10 MHN/6 MHS with heparin.
Clinical formulas of heparin as well as 4CmC induced significant higher contractures in MHS than MHN bundles.
Preservative-free heparin caused no contractures. In 13 cardiopulmonary bypass patients, no significant serum levels of 4CmC could be detected with a detection threshold of 4[micro sign]M 4CmC.

CONCLUSION: Heparin formulas, containing 4CmC, induce dose dependent contractures in vitro in MHS human skeletal muscle at about 60 [micro sign]M 4CmC.
However in vivo, 4CmC serum concentrations with therapeutic heparinization are at least 1/15 lower.
The lipophilicity of 4CmC with a high volume of distribution may account for this findings.
MHS patients seem to be not at risk by clinical heparin formulas, containing chlorocresol.

In clinical studies, 2%o p-Chloro-m-Cresol seems to be a skin irritant.
Predictive patch tests using 5% p-Chloro-m-Cresol as the challenge were negative, but provocative patch tests yielded positive responses in a few individuals.
Other data pertinent to the safety assessment of thisingredient are not available, including the concentration of use in cosmetics, UV absorption data (photosensitization data also are needed if theingredient absorbs in the UVA or UVB region), dermal developmental toxicity data and a mutagenicity study using a mammalian system (a 2-year dermal carcinogenicity study is also needed if the mutagenesis study is positive).

The aim of this study is to evaluate the characterization and disinfection efficacy of chlorocresol nanoemulsion disinfectant (CND).
The characterization of CND was examined by observing the appearance with the naked eye, identifying the structure type with dye tests and dilution tests, investigating the morphology with transmission electron microscopy, and measuring droplet size with a laser particle size analyzer.
Conductivity, viscosity and pH values were separately measured and the stability was assessed by centrifugation tests and accelerated tests.
The disinfection efficacy of CND was investigated with quantitative bactericidal tests of a bacteria suspension of E. coli 8099 and on-the-spot disinfection tests of the object surface.
The results showed that the appearance of CND was transparent and no phase separation occurred after centrifuging, the structure type was an O/W nanoemulsion, the morphology was spherical droplets and the mean droplet size was 27.43 nm with normal distribution.
The conductivity, viscosity and pH values were 479.67 μs cm−1, 204.33 cp and 2.52, respectively.
CND showed good stability and could be stored for 2 years.
In addition, CND exhibited strong bactericidal activity increasing with the increase of the disinfectant concentrations and disinfection time.
Moreover, the bactericidal ability of CND was stronger than that of chlorocresol aqueous solution (P < 0.01).
In conclusion, the results suggested that CND was an effective novel nanoemulsion disinfectant.

THE CHLOROCRESOL METHOD FOR FIELD COLLECTING:
For some years entomologists at the South Australian Museum, in Adelaide, Australia, have been using a novel method to preserve insect specimens, including butterflies, moths, and grasshoppers, as well as run-of-the-mill insect collections such as are gathered in the course of trapping with the aid of mercury vapor lamps.
Because of the many specimens to be handled, new and rapid methods of preservation were necessary.
At first the rather odorous chemical, tri-chlor-phenol, which is a substance of a dirty sugar-like appearance, was used.
This was very useful in inhibiting development of moulds in containers with freshly killed specimens.
Using refrigeration it was possible after sealing up the cans to keep them for several weeks without apparent deterioration, then take and set them up without necessity of relaxing in moist air.
When adopted by private collectors the odor of the preservation was not such as to encourage good home relations when the domestic refrigerator was used by the entomologist.
A casual shortage of tri-chlor-phenol later led Mr. R. H. FISHER to experiment by using granular crystals of Chlorocresol (name registered in the British Pharmacopeia).
This almost inodorous chemical was an instant success, revolutionised collecting techniques, and has enabled the development of a quite nove] method of preserving specimens in the field.
